Innovative bioseparation technologies employing thermoresponsive Project Details

Abstract

Biopharmaceutical advancements, such as antibody drugs, and cell therapies like stem cells, create new challenges and opportunities for cost-effective separation and purification techniques. This study examines a bioseparation strategy with poly(N-isopropylacrylamide), a polymer that has unique temperature-dependent hydrophilic and hydrophobic characteristics. The strategy employs thermoresponsive polymers to regulate the adsorption and desorption of specific target biomolecules through temperature changes. In these studies, antibody separation and purification were conducted with temperature-sensitive polymer-coated silica beads. Additionally, a polymer-coated glass slide was used to control the temperature-dependent adhesion and release of stem cells. The polymers were also used in combination with thermoresponsive polymer-modified microfibers and columns for large-scale stem cell separation and purification

of viral vectors for gene therapy. Furthermore, the separation and purification of extracellular vesicles (exosomes) were successfully achieved through the combination of affinity peptides and thermoresponsive polymers. This approach illustrates that temperature-sensitive polymers can effectively separate and purify a wide variety of biological materials, such as antibodies, cells, viral vectors, and extracellular vesicles.
