| Abstract |
A significant environmental concern is the production and disposal of conventional plastics due to the long-term plastic waste contamination in the environment. Enzymatic recycling could be a potential solution, but the majority of plastics are chemically inert and hydrophobic, posing challenges to plastic-waste degrading enzyme activity. Inspired by lytic polysaccharide monooxygenases (LPMOs) that degrade resistant polymers and biopolymers due to the presence of solvent-exposed active sites and carbohydrate-binding modules (CBMs), this paper examines the plastic-binding competence of engineered LPMOs. The theory is that, by the substitution of the CBM with one of three different surface active accessory modules that exhibit different amphipathic properties, the ability of the plastic-binding chimeric LPMO
could be modified. The ability of these chimeric LPMOs to bind specifically to plastic was tested by examination of a diverse array of synthetic polymers, including polyesters, polyamides, and polyolefins. Results indicate that the plastic-binding properties of the engineered LPMOs vary in relation to the particular polymer and can be controlled by changes in the accessory module and reaction conditions. Notably, the complete binding of certain polymer substrates indicates that specific enzymatic tools for the reduction of plastic waste are highly achievable.
