Drug Design Delivery Intracellular Bacteria Project Details
| Abstract |
Intracellular bacteria present significant therapeutic challenges by exploiting host cellular environments to evade immune responses and conventional antibiotics. These environments necessitate drug strategies capable of overcoming host cell membrane barriers, navigating complex endosomal trafficking, tolerating diverse intracellular conditions, bypassing efflux mechanisms, and countering phenotypic tolerance. While current nanotechnological advancements, including advanced carriers, prodrugs, and host-directed therapies, offer promising avenues, their focus often remains limited to facilitating cellular entry. This project explores a next-generation approach for intracellular antibacterial therapy, emphasizing subcellular targeting, the development of dual-function delivery systems, and innovative biomimetic carriers. It integrates precise intracellular pharmacokinetics/pharmacodynamics (PK/PD) assessment and leverages artificial intelligence for drug design. The objective is to transition
from merely achieving cellular entry to enabling precise spatiotemporal regulation of drug activity within infected host cells, thereby informing the development of therapeutics designed for sustained efficacy within the intracellular bacterial niche, minimizing relapse, and mitigating the emergence of antimicrobial resistance.
