Genome sequence Bacillus velezensis LZN01 Project Details

Abstract

This project builds a complete computational workflow for assembling and annotating the genome of *Bacillus velezensis* strain LZN01, a strong biocontrol agent against the fungus *Fusarium oxysporum* f. sp. niveum. The workflow uses a hybrid sequencing strategy: short‑read data from an Illumina HiSeq platform combined with long‑read data from PacBio. With the Unicycler assembly pipeline, these reads are merged to produce a finished, circular genome. After assembly, the genome is annotated to locate: * coding DNA sequences (CDSs) * ribosomal RNAs (rRNAs) * transfer RNAs (tRNAs) A phylogenomic analysis then checks that the strain is correctly classified as *B. velezensis*. The antiSMASH tool is run to find secondary‑metabolite biosynthetic gene

clusters (BGCs). The analysis highlights clusters that make known antimicrobial compounds, including: * surfactin * fengycin * bacillibactin * bacilysin To explore how the strain might produce the compound myriocin, metabolic‑pathway reconstruction is performed. This step points to several candidate genes that could be involved, such as **epsN**, **capD**, **dagK**, **glk**, and **manA**. Overall, the computational model provides a detailed reference for studying plant‑microbe interactions, biofilm formation, and plant‑growth‑promoting traits at the genome level. It offers clear, step‑by‑step guidance for M.Tech students working in agricultural biotechnology and bioinformatics.
